Les thérapies anticancéreuses reposant uniquement sur l’induction de l’apoptose des cellules tumorales montrent souvent une efficacité limitée à long terme, en raison de la capacité des cellules cancéreuses à développer des mécanismes de résistance. Afin d’améliorer la réponse thérapeutique, plusieurs approches, telles que la chimiothérapie ou la radiothérapie, sont désormais combinées aux inhibiteurs de points de contrôle immunitaire (IPCI). Ces derniers ont pour objectif de lever les mécanismes de suppression immunitaire et de stimuler l’activation des lymphocytes cytotoxiques producteurs d’interféron gamma (IFN-γ), renforçant ainsi la réponse antitumorale. De nombreuses études ont démontré que l’IFN-γ est capable de potentialiser la mort des cellules cancéreuses induite par certains agents chimiothérapeutiques. En revanche, les mécanismes moléculaires par lesquels cette cytokine agit directement sur les cellules tumorales restent encore insuffisamment caractérisés. Nos travaux préliminaires ont montré que l’IFN-γ peut, selon le contexte, soit renforcer, soit atténuer l’efficacité d’une molécule thérapeutique. Ils nous ont également éclairé quant à l’implication de protéines, connues pour leur fonction pro-mort, au niveau des phénomènes observés. Le projet de thèse consistera à approfondir ces résultats en finalisant les études déjà engagées et en identifiant les gènes cibles de l’IFN-γ responsables de la sensibilisation des cellules cancéreuses à l’étoposide. Pour cela, un criblage CRISPR-Cas9 à l’échelle du génome sera mis en œuvre. L’objectif final est d’identifier de nouveaux biomarqueurs prédictifs de la réponse aux traitements combinant immunothérapie par IPCI et chimiothérapie, afin d’optimiser la prise en charge des patients. Ce projet offrira au doctorant une formation approfondie en biologie moléculaire et cellulaire. Les travaux expérimentaux comprendront la génération de lignées cellulaires génétiquement modifiées, l’étude des mécanismes de mort cellulaire à l’aide de différentes approches (cytométrie en flux, essais colorimétriques et de luminescence), le clonage de gènes ainsi que l’analyse de l’expression génique et protéique par RT-qPCR et Western blot. To improve therapeutic outcomes, conventional treatments such as chemotherapy and radiotherapy are increasingly combined with immune checkpoint inhibitors (ICIs). These therapies aim to overcome immune suppression and stimulate the activation of interferon-gamma (IFN-γ)-producing cytotoxic lymphocytes, thereby enhancing the anti-tumor immune response. Numerous studies have demonstrated that IFN-γ can potentiate chemotherapy-induced cancer cell death. However, the molecular mechanisms by which this cytokine acts directly on tumor cells remain poorly understood. Our preliminary findings show that IFN-γ can either enhance or reduce the efficacy of anticancer drugs depending on the cellular context. These studies have also identified several pro-death proteins that appear to play a key role in these responses. The aim of this PhD project is to build on these findings by completing ongoing studies and identifying the IFN-γ target genes responsible for sensitizing cancer cells to etoposide. To achieve this, a genome-wide CRISPR-Cas9 screen will be performed. The ultimate goal is to identify novel predictive biomarkers of response to combined immune checkpoint inhibition and chemotherapy, thereby contributing to the development of more effective and personalized cancer treatments. The successful candidate will receive comprehensive training in molecular and cellular biology.
